Isolation of Pure Culture: Select individual colonies that appear to be Bacillus subtilis and streak them onto fresh agar plates to obtain isolated colonies. Repeat this process until a pure culture of Bacillus subtilis is obtained.



Subculture: Optionally, subculture the isolated colonies onto fresh agar plates or into liquid broth for further growth and maintenance of the culture.



Identification: Perform biochemical tests or molecular techniques to confirm the identity of the isolated bacteria as Bacillus subtilis. This may include tests for catalase activity, Gram staining, and molecular methods such as PCR.



Storage: Once the identity is confirmed, store the pure culture of Bacillus subtilis in suitable conditions, such as at -80°C in a freezer or in lyophilized form for long-term preservation.
