1. Needles


2. Forceps


3. Watch glass


4. Safranin


5. Glycerine

Methods:
Place distilled water in two watch glasses.
Make an oblique cut on the leaf.
Using forceps, carefully peel a section from the leaf's upper surface and place it in one watch glass with water.
Repeat for the lower surface of the leaf, placing it in the second watch glass with water.
Add a few drops of safranin solution to both watch glasses.
Using a brush, gently place cleaned glass slides over each peel one at a time.
Cut a square or rectangular piece from each peel with a blade.
Add a drop of glycerine to each slide using a dropper.
Carefully place a cover slip over the peel using a needle.
Observe each slide under the microscope and record the number of stomata found in both the upper and lower epidermis of the four-o’clock leaf.
