Endospores are highly resistant structures produced by certain bacteria, allowing them to survive harsh environmental conditions such as extreme heat, desiccation, and chemical exposure. These spores can remain dormant for hundreds or even thousands of years.
The primary stain used in spore staining is malachite green, which binds weakly to the cell wall and spore wall. Unlike vegetative cells, spores have impermeable coats that resist staining. To enhance malachite green penetration, a steaming step is employed. During this process, the spore wall becomes more permeable, allowing the dye to attach to the underlying peptidoglycan layer. Once stained, spores retain the color even after cooling.
