technical procedure—from fixation to final staining—was performed correctly. If the positive
control fails to stain as expected, the results from the test tissue are invalid, indicating a failure in
reagents or technique.
Conversely, a negative control is designed to yield a negative result, thereby proving the specificity
of the primary reaction. For staining based on chemical reactions (e.g., PAS), this can involve
omitting a key reagent, such as periodic acid in the PAS procedure. A persistent "PAS-positive"
