Abstract:
A facile synthesis of micellar "nano" indole heterocyclic anti-cancer compounds is presented. The synthesized compounds (11-
23) were thoroughly characterized using UV-VIS spectroscopy, 'H NMR, FT-IR, and mass spectrometry. The binding energies of the DNA-compound adducts ranged from
-20.08 to -23.85 kJ mol-, stabilized by hydrophobic interactions and hydrogen bonding. These compounds were found to interact with DNA by entering its minor grooves during adduct formation, with binding constants between 1.00 × 105 and 2.00 × 105 M-1. The drug-loading efficiency and content within their micellar forms were successfully recorded. Notably, compounds 11, 12, 14, and 19 demonstrated significant anticancer activities against the HepG2/C3A cell line at a micellar concentration of 670 ML mL, inhibiting cell growth by 25-50%. The drug-likeness of these nano-formulations was assessed using Lipinski's "rule of five," predicting their potential efficacy as anti-cancer agents. Collectively, compounds 11-23 show promise for further development as effective treatments for cancer.
