Peptidoglycan , also known as murein, is an essential component of the bacterial cell wall that offers mechanical support. It is a highly preserved component of both gram-positive and gram-negative envelopes. Regardless, peptidoglycan is a dense structure in gram-positive bacteria (≥10 layers), whereas it is thin (one or two layers) in gram-negative bacteria. In terms of its structure, peptidoglycan consists of glycan chains formed from N-acetylglucosamine and N-acetylmuramic acid disaccharide units; the N-acetylmuramic section connects to extensively conserved pentapeptide or tetrapeptide stems (l-alanine–d-isoglutamine–l-lysine–d-alanine–[d-alanine].
Beta-lactam antibiotics block the final stage of peptidoglycan production by acylating the transpeptidase that is responsible for linking peptides to create peptidoglycan. The targets for the activity of beta-lactam antibiotics are referred to as penicillin-binding proteins (PBPs). This binding subsequently halts the terminal transpeptidation process and triggers loss of viability and lysis, also via autolytic mechanisms within the bacterial cell
