they have delayed onset of action (2-4 weeks) as They initially stimulate 5-HT1D auto receptors → initial decrease synthesis & release of 5-HT.
Repeated administration leads to downregulation & desensitization of these auto receptors with reuptake inhibition → enhanced serotonergic neurotransmission.
